During whole-cell recording, molecules including mRNA in the cytosol will defuse into the patch pipette. We can then collect the pipette solution as soon as we finish recording. The mRNA in this solution can be reverse-transcripted to cDNA. To detect whether the neuron expresses specific proteins, we can design their primers and perform multiplex PCR. We can also examine the transcriptom of a cell and identify unique patterns of gene expression.
